It is the 22nd Century, the Third World War (2099-2126) has decimated a large part of Earth even though nuclear weapons have not been employed during the conflict. Old nations such as England, France and China have survived but have difficulty restoring their power. On the other hand, new nations and organizations such as the Sacred Republique of Mumna and the Poseidon Organization have been born on the ashes of the world resulting from the Third World War.
In this bleak future, we are introduced to ex-L.A. SWAT members Deunan Knute and Briareos Hecatonchires. Having survived the world conflict and living in a desolated city, they are found by a young woman called Hitomi who proposes them to follow her to the utopian city of Olympus which has become the world's most powerful organization and which registers the new relations in terms of politics, economics and military between new and old nations/organizations. In Olympus, they are integrated in the prestigious ESWAT (Extra Special Weapons And Tactics) organization, whose main mission is to protect Olympus from terrorist attacks and Olympus's interests in the world.
From here on, the two main protagonists encounter a great number of other characters and organizations who will be implicated in international plots that target Olympus.

ブリアレオス・ヘカトンケイレス
MAINBriareos Hecatonchires (ブリアレオス・ヘカトンケイレス, transliterated as Buriareosu Hekatonkeiresu) is one of the two protagonists of the Appleseed franchise, the other being Deunan Knute. Briareos is a trained ES.W.A.T. operative, his point man being Deunan, whose marksmanship and strategizing skills he is still unable to rival despite his cybernetic augmentations. They met in her father's LAPD SWAT unit before World War III broke out, later both serving in the same US Army platoon during the conflict.
デュナン・ナッツ
MAINDeunan Knute (デュナン・ナッツ, transliterated as Dyunan Nattsu) is one of the two protagonists of both the Appleseed manga and anime franchises. Along with her partner Briareos Hecatonchires, she is one of ES.W.A.T.'s most important members. Known for a fiery personality, and a tendency to sometimes break ranks, Deunan is still widely appreciated by her partners for her professionalism and acumen. Outside of the job she enjoys socializing with her colleagues and Bioroid relatives, is a wearer of Prada clothing, and is also a Moto-GP enthusiast, her personal mode of transport is a high-end Coscom motorbike.

SUPPORTINGHitomi (ヒトミ) is a second-generation Bioroid, Hitomi is charged with recruiting new members of ES.W.A.T. She had led the rescue mission of Deunan Knute in the Badlands, as well as aiding her getting settled in Olympus. Although her emotions are regulated, she has interest in the emotion of love, and is disturbed by hate, as expressed by the human soldiers of the ORA, especially towards the Bioroids.

SUPPORTINGYoshitsune Miyamoto (宮本 義経, Miyamoto Yoshitsune) is an engineer testing out Deunan's mecha 42 minutes into the 2003 film.
アテナ・アレイアス
SUPPORTINGAthena Areios (more commonly known just as Athena) is the political leader of the city-state of Olympus. In the OVA she holds the rank of Inspector General. By the time of Ex Machina, she is the Prime Minister.
