諸葛岳人

Takehito Morokuzu (諸葛 岳人 (もろくず たけひと) , Morokuzu Takehito), nicknamed Gakuto (ガクト, Gakuto) or Gackt, is a secondary protagonist of Prison School, and is one of the five first-year boys in attendance of all-girls school Hachimitsu Academy.

藤野清志

Kiyoshi Fujino (藤野 清志 (ふじの きよし) , Fujino Kiyoshi), nicknamed Kiyoshi (キヨシ, Kiyoshi), is the protagonist of Prison School and is one of the five first-year boys in attendance of all-girls school Hachimitsu Academy. He had a crush on Chiyo Kurihara, though he is also the object of affection for Hana Midorikawa and it's implied that he has some feelings for her too.

根津譲二

Jouji Nezu (根 (ね) 津 (づ) 譲 (じょう) 二 (じ) , Nezu Jōji), nicknamed HideJoe or Joe, is a supporting character of Prison School, and is one of the five first-year boys in attendance of all-girls school Hachimitsu Academy. His love interest is Satou, a member of the school's cooking club. Joe was originally a secondary protagonist sharing equal focus with the other boys, but he gradually became less prominent after his release from the prison.

安堂麗治

Reiji Andou (安 (あん) 堂 (どう) 麗 (れい) 治 (じ) , Andō Reiji), nicknamed Andre (アンドレ, Andore) is a secondary protagonist of Prison School, and one of the five first-year boys in attendance of all-girls school Hachimitsu Academy. He has a masochistic love interest for Meiko Shiraki, but it later develops such for Risa Bettou, the Vice-President of the official Aboveground Student Council.

若本真吾

Shingo Wakamoto (若 (わか) 本 (もと) 真 (しん) 吾 (ご) , Wakamoto Shingo), nicknamed Shingo (シンゴ, Shingo), is a supporting character of Prison School, and is one of the five first-year boys in attendance of all-girls school Hachimitsu Academy. His love interest is Anzu Yokoyama, an ex-member of the Underground Student Council who worked as Captain of the Guards. Shingo was originally a secondary protagonist sharing equal focus with the other boys, but he gradually grew less prominent after his release from the prison.

緑川花

Hana is the fourth-best karate champion in the inter-high school championships. She is a subordinate and loyal friend of Mari Kurihara, the president of the USC and she had an unrequited love interest in Kiyoshi Fujino.

白木芽衣子

Meiko is put in charge of guard duty during the two months that the boys are incarcerated in the school's Prison Block. Meiko is the subordinate and best friend of Mari Kurihara, the president of the USC.

竹ノ宮ケイト

Kate is a slightly short and slim girl with chest-length scarlet-orange hair that is tied in two low pigtails by burgundy ribbons where it is styled in drill curls with bangs swept to the left side, green eyes that has long curlicue-like eyelashes, thin eyebrows and a normal bust.

ビッチ

Bitch (ビッチ, Bitchi) is a supporting character of Prison School and a third-year student in attendance at Hachimitsu Academy. She is the cavalry mount of the Aboveground Student Council's gyaru cavalry team during the Cavalry Battle.

田中マユミ

Mayumi Tanaka (田中 マユミ, Tanaka Mayumi) is a supporting character of Prison School and a first-year student in attendance at Hachimitsu Academy. She belongs to the same class as Chiyo Kurihara and Kiyoshi Fujino, and is a member of the school's Go Club.
