Afro (アフロ, Afuro), also known as Afro Samurai (アフロサムライ, Afuro Samurai) is the main titular main anti-hero of series of the same name.

She was a young woman who lives under the pseudonym Okiku (お菊, Okiku) given to her by the Empty Seven, a violent religious organization she was once an agent of. She later died by her contractor's hands after meeting Afro later in adulthood.

ニンジャニンジャ

Ninja Ninja (ニンジャニンジャ, Ninja Ninja) is apparently the deuteragonist of the series (the main protagonist being Afro). In the official concept art he was credited as Ninja Ninjaman (ニンジャニンジャマン, Ninja Ninja-man). He is one of the most mysterious characters of the series, as he is known to suddenly appear or disappear at particular moments almost as if he was always or was never there, be it during a battle Afro is caught in, or when Afro seems to be almost completely alone.

Jinnosuke (仁之助, 仁乃助 or 仁ノ助, Jinnosuke) or just Jinno, is an major character in the Afro Samurai series. He is later known as Kuma (クマ, Kuma; literally: "bear").

ジャスティス

He is an enigmatic and power-hungry gunslinger seeking the headbands to become the only unstoppable force and the arch-nemesis of the titular protagonist, Afro, directly being the killer of the latter's father. He was the holder of the Number One Headband for most of the first anime.

The Swordmaster, often also written as Sword Master, is a fictional character from the Afro Samurai series. In the official concept art he was credited simply as Master (師範, Shihan).
