赤夜萌香

Moka Akashiya (赤夜 萌香, Akashiya Moka) is the main heroine. She is a Vampiress in Yōkai Academy and the first person Tsukune Aono meets when he sets foot in the Yōkai realm. Moka takes a liking to Tsukune, initially because of the taste of his blood (something she very quickly becomes addicted to), but later when he tries to protect her from the Outcast Ayashi, Saizou Komiya. When the Rosario Cross around her neck is removed, the seal keeping her inner nature is released. Cold and arrogant, the Inner Moka is a completely different entity from the kind and loving Moka, going so far as to specifically remind Tsukune to "be kind to the other Moka". Tsukune is the only one able to remove the Rosario Cross around Moka's neck.

白雪みぞれ

Mizore Shirayuki (白雪 みぞれ, Shirayuki Mizore) is a Yuki-Onna who becomes part of the Newspaper Club at Yōkai Academy and one of the romantic admirers of Tsukune Aono, initially stalking and remaining utterly obsessed with him.

黒乃胡夢

Kurumu Kurono (黒乃 胡夢 Kurono Kurumu) is a Succubus and a third year student at Yōkai Academy where one of her goals was to take Tsukune Aono from Moka Akashiya.

青野月音

Tsukune Aono (青野月音, Aono Tsukune), nicknamed Tsuki {月} by Kyōko Aono) is the main protagonist of the series and a former human. He is mistakenly enrolled in Yōkai Academy, a high school dedicated to teaching monsters to peacefully live alongside humans in disguise, where he meets Moka Akashiya, a Vampire who takes a mutual interest in him and the sweet flavor of his blood.

橙条瑠妃

Ruby Tōjō is a witch from the Human World. She resides at Yōkai Academy as the board chairman's aide and continues to work for him in the second manga serialization. Despite not being an official student, she receives permission to become a member of the Newspaper Club as an adviser.

仙童紫

Yukari Sendō (仙童 紫 Sendō Yukari) is a Witch and the daughter of Fujiko Sendo, her mother and her father, Tamanori Sendo and a third-year student at Yōkai Academy, and the first Witch to join the Newspaper Club, the second being Ruby Tōjō. She initially disliked Tsukune Aono, claiming that she had fallen in love with Moka Akashiya and used her magical powers to try to drive them apart. However, after both Tsukune and Moka save her from bullying classmates, she falls for him too. Her obsession with both has driven her to try to create a three-way relationship between them (though shortly after Fang Fang's introduction, she attempted to create a larger relationship involving Kurumu and Mizore as well.) Yukari has, on occasion, developed some bizarre items, often leading to funny results or far-fetched situations.

一ノ瀬珠魚

Tamao Ichinose (一ノ瀬 珠魚 Ichinose Tamao) is in charge of the Swimming Club. At first she seems gentle and sweet, but she uses her body to lure males into the swimming club and then sucks the life energy out of them. Why Tamao is allowed as the leader of the Swimming Club is unknown.

御子神典明

Tenmei Mikogami (otherwise known as The Exorcist and The Strategist) is the former headmaster of Yōkai Academy and one of the Three Dark Lords. He is said to be over 200 years old, given that the battle with Alucard happened about 200 years ago. Later, it is revealed that his true form is a Demon God Kishin; this explains why his magic is weaker than Touhou Fuhai's since he is not naturally a spellcasting monster. Revealed along with his true face, Mikogami has three Holy Locks wrapped around his torso, perhaps to seal his monster nature. He is also the one, who took Moka Akashiya in after Akasha Bloodriver sealed her to prevent Alucard from awakening.

薬丸 麻子

She first appears when Midou Kusabi failed to kill Tsukune. After wounding him with her needle finger, she took control of the doctor, Yutaka Yuji. She then attempted to use him to inject the same substance into Moka. After she smashed a vase to his head, Mako injected the substance into Moka and used her to cut Tsukune before Kurumu intervened. Feeling angry already, Kiria Yoshii told Mako they would work together. She angrily rejected his offer and took control of innocent bystanders in the infirmary, to which Kiria mentioned she has impulse issues. When Tsukune wanted to talk to Moka about the incident, he ran into Mako who used the bystanders to attack him. Moka then intervened before Mako could kill Tsukune. After Tsukune pulled off her Rosario Cross, Moka at first doesn't transform due to the substance Mako used to control her. After Tsukune talks to Moka, Moka jabs herself, transforms, and defeats Mako. (U.S. manga translation called her a "Mongrel" alluding to the mixed heritage of a Monstrel).
