赤夜萌香

Moka Akashiya (赤夜 萌香, Akashiya Moka) is the main heroine. She is a Vampiress in Yōkai Academy and the first person Tsukune Aono meets when he sets foot in the Yōkai realm. Moka takes a liking to Tsukune, initially because of the taste of his blood (something she very quickly becomes addicted to), but later when he tries to protect her from the Outcast Ayashi, Saizou Komiya. When the Rosario Cross around her neck is removed, the seal keeping her inner nature is released. Cold and arrogant, the Inner Moka is a completely different entity from the kind and loving Moka, going so far as to specifically remind Tsukune to "be kind to the other Moka". Tsukune is the only one able to remove the Rosario Cross around Moka's neck.

白雪みぞれ

Mizore Shirayuki (白雪 みぞれ, Shirayuki Mizore) is a Yuki-Onna who becomes part of the Newspaper Club at Yōkai Academy and one of the romantic admirers of Tsukune Aono, initially stalking and remaining utterly obsessed with him.

黒乃胡夢

Kurumu Kurono (黒乃 胡夢 Kurono Kurumu) is a Succubus and a third year student at Yōkai Academy where one of her goals was to take Tsukune Aono from Moka Akashiya.

青野月音

Tsukune Aono (青野月音, Aono Tsukune), nicknamed Tsuki {月} by Kyōko Aono) is the main protagonist of the series and a former human. He is mistakenly enrolled in Yōkai Academy, a high school dedicated to teaching monsters to peacefully live alongside humans in disguise, where he meets Moka Akashiya, a Vampire who takes a mutual interest in him and the sweet flavor of his blood.

仙童紫

Yukari Sendō (仙童 紫 Sendō Yukari) is a Witch and the daughter of Fujiko Sendo, her mother and her father, Tamanori Sendo and a third-year student at Yōkai Academy, and the first Witch to join the Newspaper Club, the second being Ruby Tōjō. She initially disliked Tsukune Aono, claiming that she had fallen in love with Moka Akashiya and used her magical powers to try to drive them apart. However, after both Tsukune and Moka save her from bullying classmates, she falls for him too. Her obsession with both has driven her to try to create a three-way relationship between them (though shortly after Fang Fang's introduction, she attempted to create a larger relationship involving Kurumu and Mizore as well.) Yukari has, on occasion, developed some bizarre items, often leading to funny results or far-fetched situations.

朱染心愛

Kokoa Shuzen (朱染 心愛, Shuzen Kokoa) (Koko Shuzen and Koko in the English translated manga by the Shonen Jump Manga company) is the younger half-sister of Moka Akashiya and the second sister of the Shuzen Family. She arrives as a first-year student to Yōkai Academy. Like her sister, Kokoa is also a Vampire, but her powers are not sealed. Being younger, Kokoa is not as strong as the 'sealed' Inner Moka, but uses a Transformer Bat, named Kou (also named Koumori Nazo and Kou-Buddy (Kou-buddy being the English dub version of "Kō-chan")) to make herself more formidable.

森丘銀影

Ginei Morioka (森丘 銀影, Morioka Gin'ei) is a student at Yōkai Academy who happens to be a werewolf. As a werewolf, his powers reach its highest point when the full moon shines. Under a full moon, his power rivals vampires, the werewolf's natural enemy and his speed exceeds theirs. He is good friends with Haiji Miyamoto and a loyal person as he comes to the aid of the rest of his club members. However, Gin has a bad habit of being a pervert, which has earned him numerous beatings from Kurumu Kurono. In addition to being a werewolf, Ginei is also the former club president of the Newspaper Club.

黄 芳芳

Fang Fang Huang (黄 芳芳, Won Fanfan, Chinese pinyin: Huáng Fāngfāng) (spelled Fangfang Huang in some translations) was at first an effeminate first year student who first appears in Chapter 23 of the second serialization.

橙条瑠妃

Ruby Tōjō is a witch from the Human World. She resides at Yōkai Academy as the board chairman's aide and continues to work for him in the second manga serialization. Despite not being an official student, she receives permission to become a member of the Newspaper Club as an adviser.

九曜

Kuyō (九曜, Kuyō) is the head of the Public Safety Committee at Yōkai Academy. Kuyō enforces the group's rules with a tyrannical and sadistic iron fist, going so far as to state if someone goes against him, they are against the school. He is a Yōko, an S-Class species of monster who are amongst one of the most powerful of all Yōkai who live in Japan, and they have even known to be worshiped as divine beings. Kuyō believes that yōkai and humans will always remain enemies, claiming friendship between the two only occurs in fairy tales. After Kuyō had been defeated by Tsukune, he joined Fairy Tale and became the 3rd Subdivision Leader.
